Configuring a dual drive setup

I’m going to be upgrading my iMac to support a second SSD soon but I’m not sure how to configure the dual drive setup software-wise. I’ll still be using the hard drive that it came with and the second drive will be a 250 GB Samsung 860 EVO 2.5” SATA SSD. That SSD will go (presumably) in the optical drive bay. Any suggestions of what to do hardware-wise?

Now, off to the software part. How do I configure this dual drive setup for optimal performance? Is there a guide somewhere that I could use to set up the software part once I finish the hardware part?

You don’t need to loose your optical drive! Here’s the guide to install your SSD Installing iMac Intel 27" EMC 2390 Dual HDD or SSD Drive

Once you get the SSD installed, you’ll need to run disk utility to format the drive and then just run the OS installer (I would stick with MacOS Sierra) you likely still have on your system or can re-download it from the Apps Store app. Go to the Purchased tab to locate the installer if you can’t find it.

OK, you’ve formated the drive and installed a copy of the OS onto your drive. But before you exit the installer you’ll want to migrate your user accounts as well as the apps over to your SSD. Later on you can free up your HDD of the duplicates. Your system should restart with the SSD being the boot drive, if not go into the systems preferences - Startup Disk to set it. You’re done!

Alright, so I want the OS on the SSD and I just want to use the hard drive as another storage device? Sounds simple enough, should I clone the hard drive files over to the SSD?

Also, are you sure I don't need to lose the optical drive? The dual drive goes in where the optical drive is, right?

No you don't want to clone the drive you want to migrate using the built in tool in the OS installer or if you want to do it later use the direct app Migration Assistant in your Utilities folder. Cloning apps are passé these days as the built in tools are just better!

Once done you can clean off the duplicate apps off your HDD so its just there for your data files.

I've got it working in my system, still have my optical drive working! I think you're getting confused as the SSD is mounted just behind it. Take the time to review the guide note the mounting of the drive.
