I have a iMac Intel 27" EMC 2429 with a SSD 256GB & 1TB HDD.
Do I presume that the replacement of the SSD is the same as the Dual Drive Kit install EXCEPT my system already has the cabling and the SSD behind the optical drive, so I don't need to do steps 17 - 33?
But I am guessing where the SSD would be, can you confirm?

Yup, The Apple installed SSD is a standard 2.5" SATA unit in this series.
The SSD is mounted to a frame just behind the optical drive (unlike the the double tape method 3rd parties use).
Why are you replacing it? Unless its failing I would focus on upgrading the HD if space or even speed was your issue. I would swap out the HD for a SSHD (hybrid drive). Seagate's SSHD will play nice with the built-in thermal sensor requirement (no adapter needed). If you decide to use someone else's you may need the OWC adapter cable.
Also, if you have a fusion drive config you'll need to break the setup BEFORE you can replace the drive (either) otherwise the drives ID won't be cleared from the Fusion setup so if you repurpose the drive so your Mac sees the drive again it will try to re-syncs the files to it.

I don't want to replace it just yet - it got full & I am currently moving stuff to my 1TB, but I just didn't see anything on line for replacing only adding an SSD.
If I can move my user data (which I thin I nearly have) and can move my APPs I'll be right for a while.
Many people setup a Fusion drive config and let the OS manage where the files go. As to replacing rarely is it done. As most people leverage the SSD only as the startup and App drive and use the HD for data & medial stuff if you manage the drives your self (Vs Fusion). Even still a SSHD can still add some zip .
An answer to the original question would be great, too. I don't have the second hard drive installed, and just want to replace the 256 GB SSD with a new 1TB SSD. Unclear whether that's just a direct swap or whether I need to hack in a temperature sensor as though I were replacing the HD spindle. Thanks.
With either an Apple or a 3rd party installed SSD: The SSD its self does not have a thermal sensor. Only the primary HD has the sensor which needs to be kept running for SMC services. To gain access to the SSD you'll need to take the logic board out.
